How to calculate die cast mould slide core locking force and determine the mould base thickness to lock slider

It is a stiffness problem. You need to know or assume the injection pressure, know the area of the slide, and the amount of deflection allowed at the face of the slide. That will give you the forces and the deflection allowed - or stiffness. From there determine the locking force and mould base stiffness needed to provide more stiffness than you calculated from the above. If the base and locking system are not stiff enough, the part will not be within tolerance.
